Inheritance of transgene 1Ax1 in transgenic wheat (Triticum aestivum L.) with linear transgene lacking vector backbone sequence

Abstract: To analysis identify the heritance of transgene minimal expression cassette in transgenic plant and to find the stable transgenic lines, transgenic wheat was regenerated with linear transgene 1Ax1 lacking vector backbone sequence via particle bombardment, consecutive generations of which have been investigated to explore the heredity of transgene 1Ax1 cassetts expression. The results showed that linear transgene 1Ax1 expression cassette was stably expressed and segerated at the rate of 3 to 1 in T1 generation, suggesting the functional copies of transgene 1Dx5 integrated in one locus, abiding by Mendelian genetic law. SDS-PAGE analysis indicated that parts of transgenic lines had a new HMW glutenin band of an apparent Mr lower than that of the subunit 1Dx5.
