图3 人工核酸酶技术制备基因编辑绵羊的方法示意图 ZFN、TALEN或Cas9载体主要通过显微注射或体细胞核移植递送入体细胞或受精卵中,在细胞中介导DNA双链断裂(DNA double strand break, DSB),经过细胞内同源重组修复(HDR)或非同源末端连接(NHEJ)修复可以对内源基因进行编辑或引入外源基因,获得的基因编辑细胞经过体细胞核移植(SCNT)获得后代,基因编辑胚胎则通过胚胎移植入受体中获得后代。
