Abstract: The Small-tailed Han sheep(48), Ningxia Tan sheep(121), Tan×SH (23), Poll Dorset(48), Suffolk(24) were used to study the single nucleotide polymorphisms (SNPs) of H-FABP gene by PCR-SSCP and DNA sequencing. Five nucleotide variations were found, of which G981A, A1014C, and T1019C resulted in amino acid changes of R327H, E338A, and S340P, respectively. The remaining SNPs were silentmutation. Nine genotypes (AA、BB、CC、AB、AC、BC、AD、CD、BD) were detected in primer 2, of which AA was the major genotype. Except for the Ningxia Tan sheep and Suffolk , all breeds were in accordance with the Hardy-Weinberg equilibrium. Genetic analysis indicated that polymorphism information content (PIC) was low in the Suffolk and Tan×SH , intermediate in other breeds. These results showed that the H-FABP gene had a high level of polymorphism in different sheep breeds, and could be used as a candidate gene to analyze the correlation between its polymorphism and intramuscular fat(IMF ) content in sheep. Three genotypes were detected and defined as HH、Hh、hh in primer 4, and the genotype frequency sequence was HH>Hh>hh , there were no polymorphisms
in Small-tailed Han sheep , Tan×SH and Suffolk , and polymorphism information content (PIC) was low in the Ningxia Tan sheep and Poll Dorset.All breeds were in accordance with the Hardy-Weinberg equilibrium.
