Loss of heterozygosity at chromosome 8p21-p23 in adenocarcinoma of gastric cardia

Abstract: Objective To detect loss of heterozygosity (LOH) at chromosome 8p21-p23 in adenocarcinoma of gastric cardia. Methods Laser capture microdissection (LCM) was used to obtain homogeneous cells from the tumor and adjacent normal tissues. Subsequent multiple displacement amplification (MDA) was performed for whole genome amplification. Each tumor was assessed for allelic loss using 13 microsatellite markers covering chromosome 8p21-p23. Results Higher LOH frequency (63.2%) was observed and one common deleted region of about 1Mb was defined. Conclusion The tumor suppressor gene(s) at chromosome 8p21-p23 might play an important role in the carcinogenesis process of adenocarcinoma of gastric cardia.
