摘要: 将科研前沿引入本科实验教学,是激发学生科学探究兴趣、培养其科学思维与创新能力的有效策略。转基因动物构建技术涉及多个遗传学理论环节,将其转化为实验教学内容,有助于学生深入理解遗传信息传递及基因功能实现等相关理论。然而,以脊椎动物为对象的转基因实验体系普遍受高成本、长周期等因素限制;相比之下,以果蝇为载体的转基因动物制备实验兼具教学可操作性与科研前沿性。为此,本文设计并实施了一套以转基因果蝇制备为主线的遗传学综合实验教学方案,涵盖分子克隆、显微注射、遗传筛选与荧光显微观察4个递进模块。教学过程中,通过讲授转基因表达质粒、嵌合体、生殖系传递等遗传学理论,开展显微注射、杂交筛选、表型观察等实验操作,并拓展讨论转基因哺乳动物的操作流程,有效培养了学生解决复杂问题的能力与创新性思维。

Abstract: Integrating frontline research into undergraduate laboratory classes is an effective strategy to improve teaching quality, stimulate students’ research interest, and enhance their scientific knowledge and innovative capabilities. Transgenic animal construction technology involves multiple genetic principles; designing it as an experimental teaching module helps students deepen their understanding of genetic information transmission and gene function. Transgenic experimental systems using vertebrates are constrained by high costs and long experimental periods, whereas Drosophila-based transgenic animal experiments are feasible for teaching and can be integrated with current research frontiers. Here, we designed a comprehensive genetics experiment module on transgenic Drosophila, which includes four progressive steps: molecular cloning, microinjection, genetic screening, and fluorescence microscopy. Through theoretical instruction on transgenic expression plasmids, chimeras, germline transmission, and other genetic concepts, combined with experimental operations including microinjection, cross-based screening, and phenotypic observation, as well as extended discussions on transgenic mammalian procedures, this scheme effectively cultivates students’ problem-solving abilities and innovation.
