一个新的玉米NAC类基因(ZmNAC1)的克隆与分析

摘要: NAC转录因子是近十年来新发现的具有多种生物功能的植物特异转录因子,在植物生长发育、激素调节和抵抗逆境等方面发挥着重要的作用。本研究利用电子克隆方法,结合RT-PCR分析,从玉米中克隆了一个与NAC类基因同源的全长cDNA序列,命名为ZmNAC1(GenBank登录号EU224278),该基因全长为1029bp,具有完整的开放阅读框架(ORF,26~907bp),推测编码蛋白含有293个氨基酸,分子量为32.3KD,等电点为8.65。半定量RT-PCR分析表明,ZmNAC1可以被低温、聚乙二醇(polyethylene glycol, PEG)、高盐和ABA诱导。这些结果表明,ZmNAC1可能在植物逆境反应中起作用。本研究是有关玉米NAC转录因子基因的首次报道。

Abstract: NAC transcription factors that are unique to plants are the new type transcription regulatory factors that have multiple biological functions. They play an important role in regulation of plant growth and development, hormone level and responses to various kinds stresses. One full-length cDNA encoding the NAC-like gene homologue was isolated from maize (Zea mays) by RT-PCR and named as ZmNAC1 (GenBank accession No., EU224278). Sequence analysis shows that it is 1029 bp long and contains a single open reading frame (ORF, 26~907 bp). The predicted ZmNAC1 protein has 293 amino acids with an estimated molecular mass of 32.3 KD and an isoelectric point of 8.65. RT-PCR analysis showed that the ZmNAC1expression was induced by low temperature, PEG, salt and ABA. These results suggest that ZmNAC1 may play important roles in abiotic resistance pathways. This is the first NAC-like gene reported in maize.
