The Tissue Specific Expression of Human Serum Albumin Gene Using Goat β-Casein Gene Promoter in the Mouse Tissues

摘要: 对本所构建的pcDNA3.1-β6.7hALBm表达载体行尾静脉注射直接转染哺乳期母鼠的活体组织.通过RT-PCR检测hALBm基因在小鼠几种组织中的表达来研究该构建体的组织特异性.结果:构建体在受试鼠组织中的表达显示了较好的乳腺组织特异性.说明构建具有较长5′端上游乳蛋白调控区的乳腺特异性表达载体对组织特异性表达是必需的。
Abstract:The mammary gland expression recombinant construct,pcDNA3.1-β6.7hALBm,was directly delivered into the lactating mice via tail vein injection mediated by DOTAP liposome.The expression of hALB mRNA in various mouse tissues was determined by RT-PCR method.The results showed that pcDNA3.1-β6.7hALBm presented a specificity in the mouse mammary gland expression,indicating that it was essential to construct a mammary gland expression vector with even longer 5′ upstream regulatory region of the milk protein genes for the tissue-specific expression of foreign in the transgenic animals.
